There are many threads on route to Wayanad in the travelogue section.. Route depends on which part of Wayanad you are heading to.. One through Hunsur - Kutta - Kattikulam will take you to Toalpetty, Kuruva , Manathavadi part of Wayanad. Roads are not so good in some stretch here. Gundalpet - SultanBethery - Kalpatta is longer but roads are good and will take you to the Kalapatta part of Wayanad.. Travel time is around 5 to 6 hrs..

+1 to Gundalpet - SultanBethery - Kalpatta route.
I did this route to kalpetta 6 months back.
from:bangalore,karnataka to:12.626938,77.096558 to:kalpetta, Kerala, India - Google Maps
roads were good all the way. We were lucky enough to spot elephants along the bandipur stretch scaring the crap out of every passing vehicle.

Very true. The whole stretch is beautiful. Once you cross Nanjangud, the road is a breeze. You can do 80-100 easily. Then when you are into the Waynad section of the highway, you can enjoy the lush green views all along the road. Enjoy the ride.

If you plan to start very early and a break along the BLR-MYSR section seem to early, you can break at the Coffee Day just before Gundlupet. Its aptly positioned and helps people going towards all the sections, viz South Kerala, Ooty and Waynad.

Best Route (Fastest) would be - Bangalore->Mysore->Nanjanguad->GundulPet->Muthanga->Sultan Battery->Kalpetta. Distance 300 Kms approx. Travel time 5-6 hours.
