Like earlier discussed, there is no connection whatsoever between gain setting and the number or nature of speakers connected unless you have chosen an amp which is so powerful, that using it at full potential can damage the speakers.

The gain setting will be the same as in all cases, i.e. to match the exact pre-out voltage of the head unit it is being paired with. There aren't too many exceptions to the gain settings rule except the one I mentioned above.
